In production, the role of fungi is equivalent to the seeds of crops, and its quality affects the quality and output of edible fungus products, which is related to the vital interests of hundreds of millions of mushroom farmers.
The original species of morel can be produced in September to October of the year, and the cultivated species can be produced immediately after the original species is filled in 20-30 days, and the cultivated species must be filled in the bottle at the end of October-November. In order to ensure that the bacteria will grow for one month after cultivation, and catch up with the best fruiting period for fruiting.
1. Culture medium formula and treatment:
Formula 1: 95% wheat, 3% rice husk, 1% gypsum, 1% calcium carbonate
Formula 2: 90% cottonseed hulls, 8% sawdust, 2% calcium carbonate;
Wheat: Soak the prepared wheat in cold water at about 10-20°C for 24 hours in advance, or cook it directly. The cooked wheat grains need to be checked for no white heart.
Rice husk: soak in water or 1% lime water for 24 hours, drain the water and set aside.
At the same time, nutrients such as wood chips, corn flour, and bran can be added appropriately. After pre-wetting the medium, mix it evenly to make the water content reach 65%. Add gypsum and lime to adjust the pH value to 6.5-7.5, and then bottle it.
The bottling requires consistent tightness, preferably to the shoulder of the bottle, then flatten the surface, wash away the contaminated culture medium on the outside of the bottle wall, plug it with a cotton plug, and then seal it with a plastic film.
2. Sterilization:
Autoclave at 1.5 lbs for 1 hour and atmospheric at 100°C for 4 to 6 hours. Note that the sterilization time cannot be too long or the pressure is too high, otherwise the nutrients will be destroyed.
3. Inoculation and cultivation:
After the inoculation box is fumigated and sterilized, wash the utensils by hand and inoculate in a sterile environment.
After sterilization, it needs to be cooled to below 18°C before inoculation. One mother seed can receive 5-10 bottles of original seed, and one bottle of original seed can receive 50-100 bags of cultivated seed.
Precautions
1.Sterilization temperature, during the production of strains, the original species must be sterilized by autoclaving. The original species has a wide range of propagation and a large number. Once miscellaneous bacteria are carried, it will spread to a wide range and cause heavy losses. Autoclave as much as possible, the more thorough the sterilization, the better the effect.
2.After the inoculation of the strains, transfer them to the germination room for germination in time. The bacteria-generating room requires cleanliness, temperature control and humidity control, light protection, controllable ventilation openings, and a shelf type is appropriate.
3.For the cultivation of original species and cultivars, the initial culture temperature can be carried out at 20-22°C, and when the strains germinate and grow to colonies with a diameter of 2-3 cm, lower the temperature by 1-2°C. avoid burning bacteria or strain degradation caused by temperatures exceeding 25°C. Normally, when the hyphae reaches half of the area, continue to cool down by 1-2°C until the strains are full.
4.In the middle stage of germination, pay attention to ventilate the culture room every morning to increase the oxygen content in the room; during the whole culture process, the air humidity should be kept at 55%-65% to avoid excessive humidity and cause the growth of bacteria.
