1. Strictly test the pH value of the culture material
Suitable pH is suitable for the normal growth of black fungus mycelium. After the black fungus is bagged and before sterilization, the pH value can be adjusted to 8. The pH value after sterilization will drop to between 6.5-7.5, which is normal. If the pH value after sterilization drops below 5, it will seriously affect the survival rate of black fungus mycelium. In addition, false lime phenomenon often occurs, and it is recommended that practitioners must check the pH value (acidity and alkalinity) of the culture medium.
2. Repair of sterilization facilities
High-pressure sterilization equipment mainly detects whether pipes, valves, instruments, circuits and other parts are operating normally and whether they are damaged. If so, they need to be dealt with in time.
Atmospheric pressure sterilizers mainly detect whether there are loose seals, steam leaks, and water leaks in various places, especially old sterilization pots, which need to be carefully checked.
3. Carry out pest control and sterilization work in the bacteria breeding room
Whether it is a new bacteria room or an old bacteria room, it must be strictly disinfected and treated with insecticides, and do not leave anything to chance.
4. Pay attention to testing the quality of strains
There are two main aspects: detecting survival rate and germination status; detecting whether it carries miscellaneous bacteria and pests.
5. Thoroughly clean and disinfect the vaccination environment
Vaccination facilities should be dusted in a timely manner, and the FFU laminar flow hood filter element should be replaced in a timely manner if it is damaged. If it is a sterile workshop, sterility testing is required. Ensure that the vaccination area reaches level 100 purification.
6. Strictly prevent overheating during vaccination
The inoculation operation must be performed when the internal temperature of the bacterial bag drops below 30 degrees. In particular, liquid strain production plants must pay more attention to the issue of high-temperature inoculation. Do not use a fan to blow the bacteria bag when cooling it down. Unpurified cold air may cause hidden dangers of bacterial infection.
7. Strict disinfection
Before inoculation, the outside of the strain bag should be strictly and thoroughly disinfected. During the storage and transportation of the strain, miscellaneous bacteria will inevitably adhere to the outer wall of the strain bag (bottle).
8. The stacking density should not be too high when cultivating bacteria in the bacterial chamber
It is recommended that the total space per cubic meter should not exceed 200 bags. When stacking mushroom bags with a diameter of 10 cm and a length of 22 cm, attention should be paid to retaining ventilation channels.
9. Frequently disinfect, kill insects and ventilate frequently during bacteria cultivation
Under normal circumstances, disinfection and insecticide treatment should be carried out every seven days starting from the time of entering the culture room. Treat 3-4 times in a row and reduce the dosage by half because the bacteria rods occupy half of the space. Ventilation cures all diseases. After ventilation, miscellaneous bacteria and pests can also sneak in with the wind. It is recommended to disinfect and kill insects with a small dose of medicine after each ventilation.
10. Ensure sufficient post-maturity period before opening
If the after-ripening period is insufficient, it will be difficult to reveal the primordia. Under normal circumstances, the post-ripening period of fungus varieties with different ripening periods is also different, with 10-20 days being appropriate. In principle, after the bag of mycelium is full, the culture can be continued for 10-20 days at an environment of 20-23 degrees. The purpose is to allow the hyphae to continue to fully decompose lignin, cellulose and other nutrients in the culture medium into more hyphae, accumulate more nutrients, and lay the foundation for the subsequent growth of more and better ears.
